"Two thousand years ago, the Underworld arose and attempted to take over the world. One of these formerly evil minions saw the light of justice and decided to fight for the powers of good. He saved the human world from damnation, and sealed the gateway of the Underworld for centuries. This dark knight reborn was Sparda, and his name became a legend.

"Now the Underworld threatens mankind again. The people must depend on a new champion to heed the call and save the world from damnation."
This is where it begins; two thousand years prior to the game, the Dark Knight Sparda banished Mundus to the Underworld, and locked the gate—"secretly reigning over the human world" (king of the world or something). Sparda married and had two sons, Dante and Vergil. He died, to my understanding, soon after they were born; though in the comic he dies locking Mundus in Hell, so there are conflicting ideas there. It's then stated that both Eva (Dante's human mother) and Vergil died in a personal attack. Trish states, "You're the one who lost a mother and brother to evil twenty years ago," in the first installment of the game. Eva's body is seen by Dante in the manga, which he visits frequently in his nightmares.
Dante has no recollection of Vergil until he passes by him in the manga, though Vergil seems to know perfectly well who Dante is, and makes it his duty to stalk him. In the Devil May Cry novel, Dante is orphaned with no memories, and renames himself "Tony". He doesn't remember what happened to him prior until he passes Vergil during a mission in which he is looking for a girl named Alice. Vergil seems to know exactly what happened [to their mother] and what's going on presently. After their encounter in the manga, Vergil leaves for a year, calling out for Dante in Devil May Cry 3.
Their encounter in Devil May Cry 3 includes Dante running over the newly erected Temen-ni-Guru (Temen-ni-Gru in English) searching out his brother, thinking, for the most part, that they're going to brawl and that'll be the end of it. Unfortunately Vergil's plans run deeper than that, and his true intention is to steal the amulet around Dante's neck—given to him by his late mother—and open the gate to Hell using both Dante's and his own amulet, making it whole again. Aided by a man-turned-demon by the name of Arkham, Vergil takes Dante's amulet to open Hell and inherit the "Power of Sparda", that Sparda locked within Hell.
Uncaring of Vergil's intentions, Dante follows him throughout the game, determined to brawl with him without noble cause. Arkham injures all of them when he reveals that he wished to gain the "Power of Sparda" all along, and opens Hell by spilling Dante and Vergil's blood, sacrificing the blood of his daughter, and taking the two amulets of the twins. It isn't until Mary—or "Lady", as Dante calls her—Arkham's daughter, teaches him the meaning of having a family; that no matter what, that it is important for one to take responsibility for their own family's actions. With that, Dante follows Arkham into Hell to destroy him, and is shortly joined by Vergil. Together they kill Arkham, but Vergil is determined to inherit the "Power of Sparda", Dante, determined to stop him, battles him. Vergil falls into Hell, telling his brother that it was the place that their father lived, and that he wants to stay.
It's hard to tell what Vergil wanted the power of his father for—in his endeavor for power itself, or to protect Dante. In the aftermath, Vergil loses to Mundus, and Dante starts his business, naming it "Devil May Cry". The gate of Hell has cracked open, leaving the invitation for Mundus to trickle through. This is where the first game begins.
Trish barges into Dante's office, throwing around a motorcycle; and then getting into a bit of a brawl with him (to put it lightly). As things calm down, she tells him that Mundus has been let loose, and that she needs his help to make sure that Mundus doesn't make his move to, well, take over the world. The arrive on Mallet Island (the new gateway to Hell), and Trish and Dante go in their own directions. Eventually they meet, and Dante realizes that Trish wasn't there to ask his help at all, and instead, she was there to lure him out with her incredible resemblance to his mother. She admits to taking advantage of both Dante's feelings and his need for revenge.
In a desperate act to save Dante, Trish sacrifices herself to save him. Dante is crushed, and we hear his famous: "I should have been the one to fill your soul with LIIIIIGHT" line. Together, somehow, because Trish magically reappears, they manage to defeat Mundus, and end up becoming Devil Hunters together at the end of the game.
